newlineAbstract
newlineOryza sativa serves as a staple food worldwide and it is enriched with essential phytophenolic
newlinechemical constituents which possess plenty of disease modulating features. and#947;-Oryzanol (OZ) and
newlineEllagic acid (EA) are chemically phenolic phytoconstituents having prominent anti-oxidative
newlineeffects and have potentials to modulate molecular biochemistry in beneficial way. Hence, current
newlineinvestigation was aimed to evaluate neuroprotective and cognitive enhancement effects of Oryza
newlinesativa phytophenolic constituents in STZ induced SAD/ LOAD and Aand#946; (1-42) induced EAOD in
newlineanimal models. Both these animal models mimic the pathological relevance of neurodegenerative
newlineailments as in humans. In silico experiments suggested target specificity of FA, OZ and EA for
newlineAchE and Aand#946; (1-42). Our findings suggested the safety and cytoprotective profile of Oryza sativa
newlinephytoconstituents in presence of neurotoxins in the SH-SY5Y human neuroblastoma cell line.
newlineFurthermore, antioxidative effect by DPPH assay, anti-AchE effect against AchE enzyme from
newlineElectrophorus electricus and Aand#946; (1-42) antiaggregatory effects fulfilled prime requirements of
newlineOryza sativa phytochemicals to get explored further for neurodegenerative ailments. The in vivo
newlineexperiments suggested therapeutic effects of FA, OZ and EA in STZ induced LOAD and Aand#946; (1-
newline42) induced EOAD with reference to their biochemical as well as behavioural modulatory features.
newline30 days treatment of OZ (100 mg/kg, p.o.) and EA (50 mg/kg, p.o.) in STZ (3 mg/kg i.c.v.) induced
newlineSAD showed correction of biochemical abnormalities by improvement in oxidative stress profile,
newlineretardation of pro-inflammatory markers i.e. GFAP and CRP, AchE level, and Aand#946; plaque level.
newlineTreatment of EA i.e. EA70 and EA140 (single dose of 70 and#956;M and140 and#956;M respectively, i.c.v.) in
newlineAand#946; (1-42) (10and#956;L, 1and#956;g/and#956;L, i.c.v.) treated animals signified Aand#946; (1-42) anti-aggregatory effect, Ca+
newline+/Calpain/GSK-3and#946;/CDK5 modulatory, anti-tauopathic effects. Furthermore the study signified
newlineanti-inflammatory effects through prevention of astroglial ac
