Formulation and evaluation of Nanoemulsion and self nanoemulsifying Drug delivery systems using model drugs

Abstract

Conventional drug formulations confront foremost challenges in terms of bioavailability, newlinedosage and treatment. Concomitant adverse reactions noticeably limit their therapeutic newlineefficacy. Nanoemulsion and self nanoemulsifying drug delivery systems of propofol and newlinemacitentan have been formulated for the control of these adverse effects. Studies have newlinebeen undertaken as: (i) Formulation and evaluation of multi-dose propofol nanoemulsion newlineusing statistically designed experiments; (ii) Ocular anti-inflammatory newlinepotential after topical delivery of macitentan self-nanoemulsion; (iii) Management and newlinecontrol of intraocular pressure applying macitentan hydrogel film formulation: improved newlineeffect of surfactant and cosurfactant system; (iv) Effect of surfactant on hydration and newlinestress-strain behavior of hydroxypropyl methylcellulose film formulation; (v) Effect of newlineplasticizer in presence of soluplus in film formulation for improving dissolution of newlinemacitentan. newlinePropofol nanoemulsion was formulated using in the mixture of disodium edetate, sodium newlineoleate, thioglycerol, glycerol, egg lecithin, soyabean oil and medium chain triglyceride newlineoil, and homogenization was continued at controlled temperature of 20 and#61616;C. newlineMacitentan self-nanoemulsion has been prepared with Captex 355, Croduret 40 SS, newlineKolliphor HS 15, PEG 600, Transcutol HP or ethanol (as surfactant and co-surfactants newlinesystems). Carrageenan (100 and#956;l, 3% w/v) was injected to the upper palpebral region of the newlinerabbit eye for studying the potentiality of ocular anti-inflammation after administration newlineof macitentan self-nanoemulsifying drug delivery systems (SNEDDS). newlineHydrogel forming ocular film formulation of macitentan has been prepared in newlinehydroxypropyl methylcellulose (HPMC) matrix system using different surfactant/cosurfactant newlinesystem (Croduret 40, Kolliphor HS 15, Soluplus, Transcutol HP, Captex 355, newlineviii newlineDimethyl Sulfoxide and/or Benzalkonium chloride). Intraocular pressure was monitored newlineon normotensive rabbit model after application of film in the cul-de-sac. newlineSwelling and erosion afte
