Cellular and Molecular Profiling of Dengue Virus and to Study its Circulating Multiple Serotypes in Clinical Isolates

Abstract

Dengue a flavivirus transmit infection through Aedes mosquitoes which is a major concern in India There are nearly 25000 deaths reported annually Dengue virus contains single stranded RNA and it comprise of five serotypes naming DENV1 DENV2 DENV3 DENV4 and DENV5 Each serotype causes different kind of infection due to various serotypes For this study 192 clinical samples were collected with manifestations like fever thrombocytopenia and rashes on the skin vomiting headache bleeding and pain in abdomen conjunctiva congestion and diarrhea needed to be considered for selecting the patients sample with possibility of dengue infection through various serological and molecular techniques MICROELISA was used to detect the presence of antigen in dengue infection and for molecular estimation through RNA extraction was performed by QIAGEN commercial kit USA DENV2 infection was found to be abundant among the population found with positive with dengue infection, followed by DENV3 and then with DENV1 There was no case reported for DENV4 serotype infection 49.96 percentage of the studied populations were found to be suffered from dengue infection but 44.56 percentage of the samples collected were confirmed with the dengue infection through the molecular assays NS 1 antigen with 33.3 percentage was found in abundance in the studied samples followed by NS 1 plus IgM IgM plus IgG NS 1 plus IgG and IgM with 10.4percentage 7.29 percentage 3.12 percentage and 2.08 percentage respectively During molecular testing of the samples different ct values helped in the qualitative and quantitative analysis and estimation of dengue viral samples The sample 1 with ct value 12.22 had the highest viral load among all the other samples and the sample 96 with ct value 35.79 had the lowest viral load among all the other samples used in this study The severity of the dengue infection was diagnosed through these methods and this study revealed the quick and reliable method for the detection of dengue infection newline
