Abstract
quotOBJECTIVE: The aim of the present investigation was to formulate drug (Curcumin and Doxycycline Hyclate)-loaded biomimetic collagen alginate composite nanohybrid scaffolds which have both anti-inflammatory and antibacterial activities for potential tissue regeneration in diabetic wounds.
newlineMETHODOLOGY: The compatibility between the selected drugs curcumin and doxycycline hyclate has been studies using DSC and FTIR studies. The curcumin-chitosan nanoparticles have been prepared using ionic gelation method and evaluated for particle size, zeta potential, SEM, differential scanning calorimetry and X-ray powder diffraction studies. These prepared nanoparticles are impregnated in to collagen-alginate scaffold along with doxycycline hyclate (nanohybrid scaffold). This nanohybrid scaffold is evaluated for morphology, biodegradability, biocompatibility, in vitro drug release and in vivo wound healing studies in STZ-induced diabetic rats.
newlineRESULTS: The results of compatibility studies between the curcumin and doxycycline hyclate revealed that both drugs are compatible with each other without any interactions. The DSC and XRD studies of nanoparticles confirmed that conversion of crystalline to amorphous form of curcumin. Characterization of nanohybrid scaffolds shown a porous morphology, good mechanical strength, better water uptake, biocompatibility with controlled drug release. The in vitro antibacterial studies revealed the potential antibacterial effects of nanohybrid scaffold on various microbes present in diabetic wound infections. In vivo wound healing studies of prepared scaffolds in STZ-induced diabetic rats exposed the anti-inflammatory and tissue regeneration effects of nanohybrid scaffold.
newlineCONCLUSION: The present study suggests that this synergistic combination is a promising strategy to address various pathological manifestations of diabetic wounds and have better wound healing capability.
