Cancer is the major health concern throughout the world and also it is the second newlineleading cause of death. According to our World Health Organization survey, there were newlineabout 84 death rates identified between 2005 and 2015, and in order to lower mortality newlinerates, people are now more drawn to plants and products derived from plants, especially newlinethose who have health issues like diabetes, cancer, inflammatory disorders, rheumatism, newlineliver diseases, etc. Plants from natural sources, which has been proven by many newlineresearchers to have potential phytoconsitutents were the reason behind for their curative newlineeffects by acting through various mechanism. The plant Annona muricata Linn, belongs to newlineAnnonaceae family is a miracle plant having the secondary metabolite known as newlineannonaceous acetogenin, showed anticancer properties. Curcumin is obtained from the newlinerhizomes of Curcuma longa, family of Zingiberaceae is a traditional herbal medicine used newlineby many scientists because of its potential medicinal properties. In our present study, we newlinehave focused synergistic effects of ethyl acetate extracts of leaves of Annona muricata newlineLinn, and curcumin by in-vitro study SCC-25 cell line. The results revealed its synergistic newlineresponse against the oral cancer and the IC50 value was found to be 20µg/ml, Formulation newlineof the herbal gel was carried out by varying the concentrations of plant extract and newlinecurcumin to assess their synergistic effects in SCC-25 oral cancer cell line. Curcumin and newlineAnnona muricata leaf extract both show a synergistic inhibitory effect against the SCC-25 newlinecell line in an in vitro evaluation (IC50 = 20 and#61549;g/ml). Also, we prepared a herbal gel and newlineexamined its impact on the SCC-25 cell line. We discovered that the formulation newlinecontaining 0.6% curcumin and 0.6% ethyl acetate extract from Graviola leaves had an newlineoptimum activity and produced synergistic effects. In silico studies were carried out for newlineselected 50 annonaceous acetogenin compounds based on the literatures, to evaluate their newlinebinding affinity towards oral cancer targets Akt
