Abstract
newline Serine proteinase inhibitors produced by plants, inhibiting protein digestion in
newlineherbivorous insects, can be used as one of the best ecofriendly insect management
newlinetactics. In this context, laboratory assay was conducted to identify, purify and
newlinecharacterize the potential source of serine proteinase inhibitors and to identify its
newlineefficacy on Plutella xylostella and Spodoptera frugiperda at the Department of
newlineAgricultural Entomology, Agricultural College and Research Institute, Madurai
newlineduring 2017-21.
newlineSeeds of 38 plants from nine families were selected and screened for trypsin
newlineand chymotrypsin inhibitory activity (TIA and CIA). Out of 38 seeds screened for the
newlinepresence of TIA and CIA, 20 seeds were found to possess both TIA and CIA,
newline15 seeds had only TIA. This is the new report on the presence of trypsin
newlineinhibitor in Cassia leptophylla, Clitoria ternatea alba, Parkia biglandulosa,
newlineSolanum xanthocarpum and Tectona grandis seeds. Parkinsonia aculeata,
newlinePeltophorum pterocarpum and Jatropha carcass seeds were found as source of
newlinechymotrypsin inhibitor for the first time. Tephrosia purpurea and Momordica
newlinebalsamina seeds were identified as both trypsin and chymotrypsin inhibitor source for
newlinethe first time.
newlineAmong the seeds screened, the highest mean TIA was found in the
newlineseed extracts of M. balsamina (9879 TIU g-1
newlineof seed) followed by E. variegata
newline(2737 TIU g-1
newlineof seed) and the highest CIA was recorded in E. variegata
newline(2800 CIU g-1
newlineof seed). Hence, those two proteinase inhibitors were selected and
newlinepartially purified by ammonium sulphate precipitation. After precipitation, Fraction
newline(0-60) and Fraction (30-90) were selected based on their activity from E. variegata
newlineand M. balsamina, respectively, for further purification in gel filtration
newlinechromatography using Sephadex G-75 column. Out of 35 fractions of EvPI and MbPI
newlinecollected in Gel filtration chromatography, F12 and F13 of EvPI and F17 and F18 of
