Development of herbal formulation for osteoporosis by target based approach

Abstract

Materials and methods newlineIn vitro cell culture study using MG-63 cell line and primary mesenchymal stem cells newline(MSCs) was done to screen the selected plant extracts for different targets of bone newlineremodeling, such as cell proliferation, cell differentiation, osteoclast inhibition and newlinematrix mineralization. Plant extracts which showed significant activity; in particular newlineassay was further evaluated for their in vivo study using bilateral ovariectomy-induced newlinepostmenopausal osteoporosis rat model. Selected four drugs were mixed in equal newlineproportions and extracted by hot maceration to get the combined aqueous extract (CE). newlineThree doses of the CE (100mg/Kg, 250mg/Kg and 500mg/Kg) were evaluated for newlineantiosteoporotic activity by in vivo study. Alendronate was used as positive control. newlineVegad K. L. Abstract newlineK.B.I.P.E.R. Kadi Sarva Vishwavidyalaya 2 newlineFrom the CE herbal dosage forms, herbal granules and herbal effervescent granules newlinewere prepared and evaluated. newlineResults newlineFour plant extract selected from each of the in vitro assay targeting different aspects newlinebone remodeling are aqueous extract of Curculigo orchioides for osteoblast newlineproliferation, aqueous extract of Cissus quadrangularis for osteoblast differentiation, newlineaqueous extract of Tinospora cordifolia for osteoclast inhibition and aqueous extract of newlineTerminalia arjuna in matrix mineralization for further in vivo study. Most effective dose, newlineCE 250 in the in vivo study was converted into human dose and herbal formulations, newlineherbal granules and herbal effervescent granules were prepared and evaluated for by newlineorganoleptic and flow parameters. newlineConclusion newlineAs herbal formulations were prepared from the plants extracts which are effective on newlinethe different targets of bone remodeling, they can be used as multi-targeted therapy for newlinethe treatment of osteoporosis.
