The upsurge of ruthenium-based anticancer compounds has been drawing attention in the field of anticancer drug discovery during the last two decades, which is due to the less toxicity and broad spectrum of activity of ruthenium complexes to overcome the drawbacks of platinum-based chemotherapeutic agents. The piano-stool framework of half-sandwich Ru(II)-arene complexes provide a numerous possibilities to tune the structural features of the complexes, which leads to obtaining potential anti-proliferative activity against tumor cells. It is also established that the incorporation of clinically-used drug into Ru(II)-arene moiety confers the versatility of the structure of complexes, which can able to tune their anti-proliferative activity. In the present thesis, several organometallic half-sandwich Ru(arene) complexes bearing bio-active ligands have been synthesized and characterized. Hydrophobicity, hydrogen bonding, and planarity of ligands present in complexes play a major role in effecting strong interaction with DNA and protein, by the way exhibiting cytotoxicity against cancer cells newline
