Hepatoprotective and anti oxidant activity of Stereospermum colais and Pedilanthus tithymaloides in paracetamol induced liver toxicity

dc.description.abstractIn the present study, the Stereospermum colais and Pedilanthus tithymaloides leaves were selected to evaluate hepatoprotective effect in preclinical models. The Stereospermum colais and Pedilanthus tithymaloides were evaluated for hepatoprotective activity in paracetamol and carbon tetracloride induced liver toxicity in Wistar rats. Both plants extracts were screened for preliminary phytochemical studies. Plants extracts were also evaluated for their antioxidant potential by employing in-vitro antioxidant models. The ethanolic extract of Stereospermum colais and Pedilanthus tithymaloides leaves show excellent DDPH and nitric oxide radical scavenging activity in a dose dependent manner. Stereospermum colais was found more potent as compared to Pedilanthus tithymaloidesin both in-vitro antioxidant models. Acute oral toxicity study was performed to find out the test dose of extracts as per OECD 423 guidelines and both plants extracts were found to be safe at a dose of 2000 mg/kg body weight. Hepatoprotective activity was studied against paracetamol and caron tetrachloride induced hepatotoxicity, using silymarin (25 mg/kg body weight.) as a standard reference. The biochemical parameter (such as SGPT, SGOT, ALP, total bilirubin and total protein) and histopathology of liver were also studied along with the liver homogenates (such as LPO, SOD, CAT, GSH, GPx and GST). The treatment with Stereospermum colais and Pedilanthus tithymaloides were able to restore the liver weight and biochemical parameters along with liver homogenates as compared to animals of toxic control groups. The maximum effect was seen with 400 mg/kg body weight dose of the extracts than the 200 mg/kg body weight in both hepatoprotective animal models. The ethanolic extract of Stereospermum colais leaves was found more effective than Pedilanthus tithymaloides leaves. The present investigation could also be useful to new researchers for antioxidant and hepatoprotective effects in human subjects. newline
