Optimization of Starch Degradation Microorganisms to Using Natural Resources from Selected Flour Mills of Indore City

Abstract

In this research was found the main focus on the identification of starch degrading newlinebacteria and their role in amylase production. To accomplish this task, experiments newlinewere selected and designed as per the requirement in this research. In which selection newlineof starch rich soil sites at different flour mills such as Ambikapuri, Shivampuri, and newlineVishnupuri for used abbreviation as ASV in the city of Indore district of Madhya newlinePradesh, collection of soil samples for analysis and characterization of physical, newlinechemical and biological properties were found.In soil samples has contain many newlinevaluable substrates in which included soil colour, texture, pH, organic carbon, newlinenitrogen, phosphorus and potash were important properties to be detected, for which newlinesoil samples obtained from ASV flour mills at all the three sites were analysed. newlineMicroorganisms found in soil samples obtained from ASV sites were isolated and newlineanalyzed. For which soil samples were dissolved in distilled water and the solution newlinewas filtered through used Whatman paper No. 1, the X1-10 series was prepared for newlineserial dilution of filtered samples. Selected number 10-5 test tubes from each set for newlineisolation of micro-organisms in correct density. Microorganisms were isolated from it, newlinefor which it was cultured in culture medium with starch at 37 °C for 24 h. Colonies of newlinemicroorganisms with the highest efficiency were selected and pure cultures were newlineprepared, which were used in all further experiments.Various techniques were used in newlinethe identification and characterization of isolated microorganisms, including culture newlinecharacterization in which colony size and shape were determined, Gram-stain-assisted newlineobservation under a microscope, biochemical testing of microorganisms, and newlinemolecular based estimation of genomic and plasmid DNA purity and quantity. Based newlineon the following tests, it was found that the microorganism obtained was Bacillus newlinespecies of bacterial group.The obtained bacteria were used for starch degradation and newlinedetermination of maximum yield production of amylase
