dc.description.abstractnewline Abstract: newlineIntroduction: To achieve targeted drug delivery to the brain, researchers are continuously involved in developing and evaluating a wide variety of drug molecules and their derivatives against Alzheimer s Disease. But there are some drawbacks associated with current therapies which can be solved by nanoformulation such as nanoemulsion. Objective: The objectives of the proposed research work were to design and develop an effective herbal nanoformulation for the treatment of Alzheimer s. Curcumin (Cur) and piperine (Pip)are very well-known phytochemicals that are claimed to have many health benefits and have been widely used in foods and traditional medicines. This study investigated the therapeutic efficacy of these compounds to treat Alzheimer s disease (AD). However, poor oral bioavailability and permeability of curcumin is a major challenge for formulation development scientists. In this research study, the researcher tried to enhance the bioavailability and permeability of curcumin with a nanotechnological approach. Objective of present study was to design, and prepare a CUR-PIP loaded solid- SNEDDS (self-nano emulsifying drug delivery system) to enhance their solubility, bioavailability, permeability, and evaluate its therapeutic effectiveness on AD animal model in comparison with untreated AD model and market formulation (donepezil). Further convert this formulation it into a proper unit dosage form. Hypothesis: Curcumin has low bioavailability but when curcumin is administered along with piperine, it increases the bioavailability of curcumin by 154% in rats and 2000% in human. The blood brain barrier (BBB) prevents the brain to take most of the pharmaceuticals from blood circulation.
