dc.description.abstractBackground newlineEndodontic treatment is the treatment of choice in apical periodontitis as the newlineperiapical tissues present with inflammation and/or infection. The success of newlineendodontic treatment depends on the complete debridement of microbes newlinefrom the root canal system and prevention of reinfection. Use of irrigants and newlineintracanal medicaments to control the growth of microbes within the root newlinecanal system is thus imperative. newlineAim newlineThe purpose of this study is to evaluate the efficiency of two intracanal newlinemedicaments (Calcium hydroxide and Farnesol) against microorganisms in newlineinfected root canals by estimation of total microbial count using newlineRT-PCR/qPCR before starting the treatment and after the tooth becomes newlineasymptomatic. newlineMaterials and Methods newlineIn a prospective randomized clinical trial, 32 patients were selected with age newlinebetween 18 to 50 years according to inclusion and exclusion criteria and newlinepreoperative diagnosis, pain scores were recorded and root canal samples newlinewere collected. Based on block randomisation patients were divided into newline2 newlineGroup I :Calcium Hydroxide and Group II : Farnesol. After routine root canal newlinepreparation, in Group I and Group II cleaning and shaping was done newlinefollowing the standard protocol until 6% 25 Protaper Gold rotary files. A newlinesecond sample from the cleaned and shaped root canal was collected. newlineIntracanal medicament was placed based on the assigned group using newlinelentulospirals and the patient was recalled after seven days. Pain scores newlinewere recorded and a third sample from the root canals was obtained and newlineobturation was completed. The microbial load in the root canal samples newlinebefore and after the treatment was evaluated by RT-PCR/qPCR and newlineanalysed statistically. newlineResults newlineBoth the groups showed a decrease in the total microbial count before the newlinetreatment, after cleaning and shaping was performed and after the newlinemedicament was placed in the canals for 7 days. There was a significant newlinedifference in values of before the start of the treatment and after the newlinecompletion of the treatment in both the groups.
