dc.description.abstractCanthium coromandelicum (Burm. f.) Alston is a medicinal plant belonging to family of Rubiaceae. This plant is used in the treatment of scabies, ringworm, cough, indigestion and snake bite. It shows various pharmacological properties like antioxidant, preliminary anti-inflammatory, antirhuematic, wound healing and diuretic activity. newlineIn the present work, protocols for in vitro seed germination, direct /indirect shoot regeneration and in vitro production of normal root culture were developed. The research work was also carried out to evaluate anti-diabetic, anti-inflammatory and antimicrobial activity of the selected plant. newlineHigh frequency in vitro clonal propagation protocols were standardized from the nodal explants derived from in vitro raised 30 days old seedlings. Knudson C (KC) medium was the best medium for seed germination and seedling development. While, maximum shoot regeneration frequency through callus was obtained on MS (Murashige and Skoog medium)+ 4 mg/l BAP (6-benzyl amino purine)+ 1 mg/l IAA (Indole -3- acetic acid), optimum direct shoot multiplication frequency was obtained on MS (Murashige and Skoog medium)+ 3 mg/l Kn ( Kinetin)+ 1 mg/l IAA (Indole -3- acetic acid). The elongated shoots were sub cultured on ¼ MS with 3 mg/l NAA for rooting. The in vitro raised plantlets were acclimatized in green house and progressively transplanted to natural conditions with 70% survival. newlineOptimum result of fast growing normal root culture using leaf explants under total darkness was obtained on solid ½ MS+2.5 mg/l IBA+1 mg/l NAA with 14.30±1.08 number of roots per culture with 3.00±0.42 gm fresh biomass per culture. In liquid culture, there was maximum of 1.91±0.05 gm dry biomass after fifth week in same medium. In vitro produced fast growing roots showed similar chemical profile like that of natural roots. newlineThe present work was also carried out to evaluate anti-inflammatory and anti-diabetic activity of different crude leaf extracts of Canthium coromandelicum (Burm. f.)
