dc.description.abstractAbstract newlineThe effectiveness and safety of Ayurvedic formulations are highly dependent on their preparation methods, particularly in ensuring standardized quality and therapeutic outcomes. This study focuses on the preparation and evaluation of Arkadiyog (AY) and its Modified form as Avaleha as Arkadiyog avaleha (AYA). A systematic approach was adopted to assess the pharmaceutical, analytical, and preclinical aspects of these formulations. newlineThe research involved authentication and analysis of raw materials, followed by the preparation of AY as per classical text mentioned in Rasaratnasamuchhya as Kasahara yog. Analytical studies were conducted to evaluate physicochemical properties and chromatographic profiles. In this study AY and AYA were prepared analyzed and assessed for anti-inflammatory and anti-allergic activity. A comparative preclinical evaluation was performed on Wistar Rats. The experimental study (In-vivo) was carried as Acute Toxicity Study, Anti-inflammatory study as Acute inflammation - Carrageenan induced paw oedema (CIPM) model and for Sub-acute inflammation- Cotton pellet granuloma model (CPGM). The Anti-Allergic Study was performed by Mast Cell Degranulation Method (MCDM). newlineIn acute toxicity study of AY and AYA in Wistar rats as per the OECD guideline 423, results showed that the test drug did not cause mortality in the treated rats at the higher dose of 2000 mg/kg body weight. AY and AYA when tested for Acute inflammatory study by CIPM show positive result. As comparative to AYA Test 2 group, Test 1 group AY shows better result. Similarly, both test drug shows positive result in CPGM. In this cotton pellet granuloma method for sub-acute inflammation AY shows better result than AYA. Both test drug proves its anti-allergic effect in MCDM. newlineThe experimental study in Wistar rats has generated valuable data on Acute toxicity study, Anti-inflammatory activity and Anti-allergic activity of Arkadiyog and Arkadiyog Avaleha, contributing to its potential therapeutic efficacy Kasahara. newline
