dc.description.abstractThe present thesis deals with the investigations carried out by the writer on development and evaluation of oral controlled release formulations namely tablets and spheroids and buccoadhesive tablets with the objective of evaluating a novel polysaccharide and improving the pharmacotherapy associated with the selected drug candidates. newlineThe trends in drug delivery and introduction to oral controlled release and buccal drug delivery systems. A brief note advantages of oral controlled release and buccal route of administration over the conventional delivery, method of manufacture and polymeric systems used for the development with special emphasis on the hydrogels is presented. Literature review of the prior art in these fields and the current developments for the selected drug candidates is being mentioned. newlineThe plan of work is presented in the second chapter of the thesis. The importance of the hydrogels in drug delivery and need for the development of novel polysaccharide hydrogels is being emphasized. The materials and the experimental techniques used for the isolation, characterization and toxicity studies for the novel polysaccharide, preformulation studies, development and evaluation of oral modified release tablets, spheroids and bilayered buccal delivery systems, stability studies, bioanalytical methods used for the estimation of the selected drugs in plasms and its validation and bioavailability studies. newlineIt concluded that the isolated polysaccaride hydrogel from Prunus amygdalus can be used as a release modifier in the development of controlled release drug delivery systems and as a mucoadhesive component for the development of transmucosal delivery systems. The developed controlled release tablets, spheroids and buccoadhesive tablets for the selected drug candidates can overcome the drawbacks of the conventional drug therapy and can improve the pharmacotherapy. newline
