Abstract
Gastric cancer is the major health problem in both men and female No efficient biomarker
newlineis available to characterize the mechanism of the cancer especially with regards to
newlinemetastasis Searching for potential gene is a key to understand the molecular mechanism
newlinebehind the gastric cancer development In this study I applied a comprehensive strategy
newline to identify miRNA by searching human EST sequences involved in gastric cancer
newlineagainst known mature miRNA sequences and to predict its target protein I also identified
newline the expression of miRNA by Real Time PCR and target protein expression
newline by immunohistochemical technique in both gastric cancer and adjacent normal tissues
newlineThe expression of miR564 was down regulated in gastric cancer tissues compared
newlineto the adjacent normal gastric tissues Target protein RelA and IL6R expressions
newlinesignificantly increased in gastric cancer tissues The expression of both miRNA
newlineand targets proteins were negatively correlated which indicated that miR564 might
newlinebe a novel negative regulator of RelA and IL6R proteins and plays a vital role in
newlinetumorigenesis of gastric cancer The expression of miR564 was significantly correlated
newline with the depth of tumour invasion suggest that miR-654 may be involved in the progression
newline of gastric cancer This data reveals that miR-564 acts as a potential tumour suppressor and
newline miR564 RelA IL6R complex may be a novel prognostic and therapeutic target for
newlinegastric cancer treatment
